Purpose Protein L Agarose is an affinity chromatography medium designed for easy, one-step purification of classes, subclasses and fragments of immunoglobulins.
Specificity Protein L only binds to Ig containing kappa light chains type kappa I, III, and IV in human, kappa I in mouse and specific for certain kappa subgroups in other species. Protein L binds weakly to rabbit Ig and does not bind Ig from bovine, goat, sheep or Ig with lambda light chains. Protein L is not appropriate for general polyclonal antibody purification from serum, which contains a mixture of immunoglobulins containing different types of light chains.

Background Protein L is first isolated from the surface of bacterial species Peptostreptoccus magnus and now recombinant Protein L is produced. Unlike Protein A and Protein G, which bind primarily through Fc regions of immunoglobulins Protein L binds immunoglobulin kappa light chain and binds a wider range of Ig classes and subclasses. Since no part of the heavy chain is involved in the binding interaction, Protein L can bind to all classes of Ig (i.e., IgG, IgM, IgA, IgE, and IgD) and also the single chain variable fragments (Scfv) and Fab fragments.
