Purpose

The ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utlizes monoclonal antibody specific for MEP1A coated on a 96-well plate. Standards and samples are added to the wells,and any MEP1A present binds to the immobilized antibody. The wells are washed and a horseradish peroxidaseconjugated rabbit anti-MEP1A monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich".The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount ofMEP1A present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of themicrowell are read at 450 nm.

Sensitivity

The minimum detectable dose of Mouse MEP1A was determined to be approximately 78.125 pg/mL. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

Capture Antibody - 1.0 mg/mL of rabbit anti-mouse MEP1A monoclonal antibody. Dilute to aworking concentration of 2.0 μg/mL in CBS before coating. Detection Antibody - 0.25 mg/mL rabbit anti-mouse MEP1A polyclonal antibody conjugated tohorseradish-peroxidase (HRP) . Dilute to working concentration of 0.5 μg/mL in detection antibodydiluteion buffer before use. Standard - Each vial contains 170 ng of recombinant mouse MEP1A. Reconstitute standardpowder with 1mL detection antibody dilution buffer. After reconstitution, store at -20 °C to -70 °C in amanual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilutionbuffer, and a high standard of 5000 pg/mL is recommended.
